FORMULATION AND CHARACTERIZATION OF ATORVASTATIN SOLID DISPERSIONS FOR IMPROVED BIOAVAILABILITY

Authors: Manju Singh, Richa Sharma, Vijay Kumar*

Solid dispersions (SDs) of atorvastatin (ATS) using PEO-PPO block copolymer resulted in the formation of an amorphous product with enhanced dissolution and bioavailability. Solubility enhancement was observed in the presence of selected carriers and it increased with the increase in concentration of carriers. The carrier and the active ingredient are dissolved in a suitable organic solvent is evaporated to an elevated temperature or under vaccum. As the solvent is being removed, super saturation occurs followed by simultaneous precipitation of the constituents resulting in a solid residue. The solvent-based process uses organic solvent to dissolve and intimately disperse the drug and carrier molecule The accumulative solubility of atorvastatin (ATS) from PEO- PPO block copolymer (Poloxamer 188) was found to be superior from glyceryl monostearate and PEG 4000 carriers due to increased content which played major role in solubility enhancement thus improving dissolution rate of SD2 batch with maximum drug release (98.08 %) in 120 minutes. The optimized SDs were characterized by saturation solubility study, drug content, in vitro dissolution, fourier transform infra-red spectroscopy, scanning electron microscopy, and x-ray diffraction. Capsules containing optimized SDs were prepared and compared with marketed brand (LIPITOR®). Finally it can be concluded that optimized SDs of ATS ameliorate the solubility and dissolution of drug. Since the batch SD2 elicited superior results, it can be proposed as good candidate for systemic product development.

A REVIEW ON CONTROVERSIES AND CLINICAL ASPECTS OF PARIBHASHA SHARIR IN RACHANA SHARIR VIGNYAN

Authors: *Dr. Vidya Deore, Dr. Avinash Deore

Sharir Rachana is an integrative branch of Ayurveda that studies the human body in its most minute form to an organized structural form. Sushruta, first carried out a detailed cadaveric study of the human body around 2000 yrs ago and laid the bedrock of Sharir Rachana Vigyan in his Samhita. Cadaveric studies thus helped the ancient seers explore many structures of human body and thus with the passing time this scientific branch evolved with a complex nomenclature of the various body parts. These nomenclatures and definitions of the basic bodily structures are compiled under the heading of Paribhasha Sharir. The knowledge of Paribhasha Sharir holds utmost importance in the field of surgery and medicine as well. However lack of social and technological advancements in the era when Ayurveda was encrypted by the Sages, there emerged out various controversies over a particular structure. Acharyas have stated out individual opinions over a single structure that created a big confusion to the forth coming generations. With developments in recent times these confusions were gradually resolved keeping the original texts and definitions unaltered. Thus the present article is a framework that reviews the controversies and applied aspects of Paribhasha Sharir in Rachana Sharir Vigyan.

FORMULATION AND EVALUATION OF MELPHALAN LOADED NIOSOME FOR CANCER TREATMENT

Authors: Dr. M. Gopal Rao*, Dr. K. Muthusamy, Dr. T. K. Ravi, Mr. Y. Arul Packiadhas, Mr. K. Jayaseelan

Cancer treatment with conventional chemotherapy drugs often faces challenges such as poor solubility, rapid metabolism, systemic toxicity, and non-specific distribution. Melphalan, an alkylating agent widely used in multiple myeloma and other cancers, is clinically effective but suffers from reduced stability and limited bioavailability. To address these drawbacks, the present work focused on developing melphalan-loaded niosomes as a novel nanocarrier system. The formulations were prepared using the thin film hydration technique with cholesterol and non-ionic surfactants. They were further analyzed for particle size, surface charge, drug entrapment efficiency, pH stability, compatibility studies, and in vitro release behavior. The optimized niosomal formulation exhibited nanoscale vesicles with uniform distribution, high entrapment efficiency, and stable zeta potential. FTIR analysis confirmed no significant drug–excipient interactions. The release profile demonstrated sustained drug release over an extended period, with kinetics indicating a diffusion-controlled mechanism. The findings suggest that melphalan-loaded niosomes can enhance drug stability, provide controlled delivery, reduce systemic side effects, and thereby improve therapeutic efficacy in cancer management.
